Биназа индуцирует провоспалительный иммунный ответ макрофагов

Аннотация
Биофармацевтические агенты, особенно белковой природы, могут обладать широким спектром иммунологических эффектов. Цитотоксические рибонуклеазы, к которым относится и биназа - РНКаза Bacillus pumilus - обладают весомым потенциалом для использования в качестве средств терапии злокачественных новообразований. Однако без всестороннего анализа реакций иммунной системы на препарат его практическое использование затруднено. С помощью проточной цитометрии оценивали качественные и количественные изменения в синтезе периферическими макрофагами мыши цитокинов IL-6, IL-10, МСР-1 и TNF-a под действием биназы и РНКазы А. Установлено, что в исследованных концентрациях (100 и 300 мкг/мл) биназа, в отличие от РНКазы А, способна активировать провоспалительный ответ макрофагов, индуцируя повышение в среде культивирования уровня цитокинов IL-6, МСР-1 и TNF-a. Обработка РНКазами не приводила к усилению синтеза антивоспалительного цитокина IL-10. Полученные данные позволяют предположить, что способность биназы активировать провоспалительный ответ макрофагов усилит ее противоопухолевое действие при применении in vivo .
