Consequently, searching for new effective methods of its treatment is now gaining popularity. The most promising approach is transplantation of tissue-engineered constructs consisting of cultured limbal stem cells (LSCs) and variety of biopolymer carriers.</p> <p><bold><italic>AIM</italic></bold><italic>: </italic>This study was performed to obtain and characterize a tissue-engineered constructs consisting of cultured LSCs and collagen membrane.</p> <p><bold><italic>MATERIALS AND METODS</italic></bold><italic>: </italic>The study was performed at the Krasnov Research Institute at the Krasnov Research Institute of Eye Diseases and Koltzov Institute of Developmental Biology in cooperation with IMTEK Ltd. with a series of experiments. Two Chinchilla rabbits with an average weight of 3.5 kg and age of 6 months have been involved in this trial. LSCs were isolated and cultured in vitro from the healthy eye of rabbits using a method modified by the authors. The abtained cells were then cultured for 14 days and transplanted to the collagen membrane, which was then examined using immunohistochemical analysis.</p> <p><bold><italic>RESULTS</italic></bold><italic>:</italic> The cells isolated from the biopsy were a mixture of fibroblast-type cells and cells with characteristics of LSC. They maintained high survivability, proliferativity, phenotype and stemness on the collagen carrier according to immunofluorescent study.</p> <p><bold><italic>CONCLUSIONS</italic></bold><italic>:</italic> Thus, the abstained tissue-engineered constructs could be used for further transplantation to the affected eye with limbal stem cell deficiency under experimental conditions.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Обоснование</bold>. Лимбальная недостаточность является трудноизлечимой патологией, при которой консервативные методы не всегда эффективны, а хирургия лимитирована количеством и доступностью тканей. В связи с этим важен поиск альтернативных методов лечения. На сегодняшний день наиболее перспективным подходом является трансплантация тканеинженерных конструкций, состоящих из культивированных лимбальных стволовых клеток (ЛСК) в составе биополимерных носителей.</p> <p><bold>Цель исследования</bold> — получить и охарактеризовать тканеинженерную конструкцию из культивированных ЛСК и коллагеновой мембраны.</p> <p><bold>Материалы и методы</bold>. Последовательная серия экспериментов выполнена на базе НИИ глазных болезней им. М.М. Краснова и Института биологии развития им. Н.К. Кольцова в сотрудничестве с ООО «ИМТЕК». В качестве источника стволовых клеток лимбальной зоны роговицы использованы 2 кролика породы шиншилла (средний возраст — 6 мес, средняя масса тела — 3,5 кг). Стволовые клетки выделены и выращены <italic>in vitro</italic> модифицированным авторами способом. Полученные клетки культивировали в течение 14 дней и пересаживали на коллагеновую мембрану, которую затем исследовали с помощью методов иммуногистохимии.</p> <p><bold>Результаты</bold>. Клетки, выделенные из биоптата, представляли собой смесь клеток фибробластного типа и клеток с характеристиками ЛСК. Они сохраняли высокую выживаемость, пролиферативную активность, фенотип и стволовость на коллагеновом носителе.</p> <p><bold>Заключение</bold>.
