CXCR4 is known to be an indirect target for G-CSF stimulation. The aim of this study was to evaluate the level ofCXCR4 expression on the surface of HSC, collected from apheresis products of patients with different responses to G- CSF-stimulation. Cord blood, normal bone marrow and primed bone marrow samples were analyzed to be compared with apheresis product. It was demonstrated that mean fluorescence intensity of CXCR4 indicating the density of CXCR4 expression on HSC surface is diminished after G-CSF stimulation towards normal bone marrow ? primed bone marrow ? apheresis product from mobilizable patients and demonstrates negative correlation with mobilization efficacy. CXCR4 expression on the surface of HSC from non-mobilizable patients does not differ from other native sources, such as normal bone marrow and cord blood, that points out the intrinsic unresponsiveness to G- CSF treatment or other kinetics of mobilization in poor-mobilizable patients.</p></abstract><trans-abstract xml:lang="ru"><p>CXCR4 является хемокиновым рецептором, который играет ключевую роль в регуляции процессов направленной миграции гемопоэтических стволовых клеток (ГСК). CXCR4 представляет собой непрямую мишень для действия Г-КСФ. Целью данной работы было проведение сравнительного анализа экспрессии CXCR4 на поверхности CD34+ ГСК, полученных из продукта афереза мобилизованной периферической крови пациентов с различной эффективностью мобилизации. В качестве групп сопоставления в исследование включены также образцы праймированного костного мозга, нормального костного мозга и пуповинной крови. Было выявлено, что средняя интенсивность флюоресценции CXCR4, отражающая плотность экспрессии данного рецептора, снижается при стимуляции Г-КСФ в направлении: нормальный костный мозг &gt; праймированный костный мозг &gt; продукт афереза от мобилизуемых пациентов и негативно коррелирует с успешностью мобилизации. Уровень экспрессии CXCR4 в группе немобилизуемых пациентов не отличается от нативных источников (костный мозг, пуповинная кровь), что указывает на внутреннюю резистентность к действию Г-КСФ или иную кинетику мобилизации у пациентов с плохим ответом на мобилизацию.</p></trans-abstract><kwd-group xml:lang="en"><kwd>CXCR4</kwd><kwd>CD34+ hematopoietic stem cells</kwd><kwd>mobilization</kwd></kwd-group><kwd-group xml:lang="ru"><kwd>CXCR4</kwd><kwd>гемопоэтические стволовые клетки</kwd><kwd>мобилизация</kwd></kwd-group><funding-group/></article-meta></front><body></body><back><ref-list><ref id="B1"><label>1.</label><mixed-citation>Copelan E.A.
