It was shown previously that well-known methods of decellularisation including treatments with enzymes or some detergents or with hypotonic solution didn't suppress calcification of implanted aorta. In the present study we showed potential in prevention of calcinosis by a treatment, which induced death of donor cells without their total destruction before implantation. This method of anticalcinosis devitalisation is based on a hypothesis of mechanism of calcinosis initiation in heart valve transplants, which was earllier proposed in our group.</p></abstract><trans-abstract xml:lang="ru"><p>Подавление кальциноза и дегенерации трансплантатов клапанов сердца остается важной задачей сердечно-сосудистой хирургии и тканевой инженерии, решение которой необходимо для увеличения сроков функционирования биопротезов. Ранее нами было показано, что предложенные в литературе способы децеллюриларизации трансплантатов путем ферментативной обработки, осмотическим шоком либо с помощью детергента додецилсулфата натрия не предотвращали кальциноз стенок аорты. В настоящей работе, используя модель подкожной имплантации тканей крысам линии Wistar, мы показали возможность полного подавления кальцификации ксенографтов клапанов сердца путем их инкубации до имплантации в растворе с дигитонином и ЭДТА, вызывающей гибель клеток донора без разрушения хроматина.
